Abstract: Periodontitis is a chronic inflammatory disease characterized by progressive destruction of the periodontal ligament, alveolar bone, and surrounding soft tissues, ultimately leading to tooth loss if left untreated. Conventional periodontal therapies primarily focus on infection control and defect stabilization but are limited in their ability to achieve complete and predictable regeneration of periodontal tissues. Therefore, there exists a significant need for advanced biomaterials capable of promoting truetissue regeneration-while providing_localized“therapeutic"actioiL The present invention discloses a bioactive hydrogel composition incorporating oleuropein and horsetail (Equisetum spp.) plant extract for periodontal regeneration. The hydrogel serves as a three-dimensional scaffold that mimics the natural extracellular matrix, facilitating cell adhesion, proliferation, and differentiation at the periodontal defect site. The hydrogel matrix enables controlled and sustained release of the incorporated phytochemicals, thereby maintaining prolonged therapeutic activity. Oleuropein provides potent anti-inflammatory, antioxidant, and antimicrobial effects, reducing periodontal inflammation and inhibiting pathogenic bacteria associated with periodontitis. Horsetail plant extract, rich in silica and flavonoids, enhances collagen synthesis, angiogenesis, and osteogenic activity, contributing to regeneration of periodontal ligament and alveolar bone. The synergistic combination within the hydrogel promotes accelerated wound healing, improved tissue integration, and enhanced structural regeneration. Accordingly, the disclosed hydrogel composition offers a biocompatible, minimally invasive, and effective regenerative approach for the treatment of periodontitis, overcoming limitations of existing periodontal therapies and providing improved clinical outcomes.
