Abstract: The present invention relates to a bioinspired peptide-functionalized Efinaconazole-loaded nanostructured lipid carrier gel for enhanced transungual delivery and a process for preparation thereof. The gel comprises Efinaconazole-loaded nanostructured lipid carriers, comprising: Compritol 888 ATO as a solid lipid, Capryol 90 as a liquid lipid, Tween 80 as a surfactant, and a decorin-derived keratin-binding peptide having the amino acid sequence LRELHLNNC associated with the surface of the nanostructured lipid carriers, dispersed in a pharmaceutically acceptable gel base. The nanostructured lipid carriers are prepared by hot homogenization followed by probe ultrasonication and subsequently functionalized with the keratin-binding peptide to enhance interaction with nail keratin. Formulation variables including solid lipid concentration, liquid lipid concentration, surfactant concentration and sonication time are optimized using a Box–Behnken Design. The optimized peptide- functionalized nanostructured lipid carrier gel exhibits an entrapment efficiency of about 87.3±1.4%, sustained drug release of about 72% at 24 hours and about 80% at 72 hours, nail penetration depth of about 1.21±0.10 mm, and a zone of inhibition of about 28.9±1.91 mm against Candida albicans. The gel provides enhanced nail retention, improved transungual penetration, sustained antifungal activity and localized delivery of Efinaconazole for management of onychomycosis.
