Abstract: The present invention discloses a biology-constrained temporal generative multimodal graph-transformer system and method for risk stratification and clinical decision support in hematologic malignancies. The system receives heterogeneous biomedical data comprising multi-omics, whole-slide pathology images, clinical narrative data, flow cytometry, laboratory information, and electronic health records. The data are transformed into unified token representations, integrated through a heterogeneous biological interaction graph using relation-aware graph reasoning, and processed by a temporal transformer to model longitudinal disease progression. A probabilistic missing-modality inference framework estimates latent representations for incomplete observations and generates uncertainty-aware predictions. A multi-task prediction engine produces disease risk, molecular subtype, relapse, survival, and prognosis predictions, followed by probability calibration to improve predictive reliability under incomplete multimodal clinical data.
