MUMBAI, India, Aug. 17 -- Intellectual Property India has published a patent application (202541011759 A) filed by Kcat Enzymatic Private Limited on February 12, 2025, for Biosynthesis Of Paracetamol Using Metabolically Engineered E. Coli.

Abstract: BIOSYNTHESIS OF PARACETAMOL USING METABOLICALLY ENGINEERED E. COLI. This invention provides a green route via a metabolically engineered E. coli for paracetamol synthesis from glucose as the sole carbon source. The synthetic designed pathway uses tailored shikimate and glycolysis pathways for improved chorismate and acetic acid flux, respectively. The tailored shikimate pathway uses engineered genes to convert glucose to paracetamol, including a gene with a feedback resistance mutation for aromatic amino acid. Additionally, genes for the improved production of chorismate from shikimate were introduced into the metabolically engineered E. coli cell and genes from the aromatic amino acid synthesis pathway, folate pathway and electron transporter synthesis pathway were downregulated to ensure high titre of intermediates for paracetamol production. The penultimate step, converting p-aminophenol to paracetamol, requires acetic acid from the tailored glycolysis pathway. This engineered E. coli, utilizing synthetic pathways for chorismate and acetic acid, produces 10 g/L of paracetamol from 200 g/L glucose loading.
