Abstract: ABSTRACT Title: Brain-Directed Functionalized Biodegradable PLGA Nano-Delivery System for Donanemab in Neurodegenerative Disease Management The present invention relates to a transferrin-functionalized biodegradable nanomaterial system encapsulating donanemab for neurological therapeutic applications, particularly Alzheimer’s disease. The invention comprises a triethylene glycol (TEG)-gelatin nanogel encapsulating donanemab, further incorporated within poly(lactic- co-glycolic acid) (PLGA) nanoparticles and surface-functionalized with transferrin for facilitating interaction with transferrin receptors associated with the blood–brain barrier. The developed nanomaterial exhibits nanoscale particle size, high encapsulation efficiency, colloidal stability, and sustained drug release characteristics. The TEG-gelatin nanogel core provides a hydrophilic environment suitable for monoclonal antibody incorporation during nanoparticle fabrication, while the PLGA matrix enables controlled and prolonged release of donanemab. Surface functionalization with transferrin provides a receptor-associated nanoparticle platform for neurological delivery applications. The invention provides a biocompatible and sustained-release monoclonal antibody nanocarrier system with potential utility in the therapeutic management of Alzheimer’s disease and related neurodegenerative disorders.
