Abstract: The present invention relates to a chemotherapeutic composition for oral administration comprising lipid-polymer hybrid nanoparticles (LPNs) encapsulating a Biopharmaceutics Classification System (BCS) II/IV histone deacetylase inhibitor (HDACi) drug and a blend of equivalent amounts of block co-polymer, solid lipid, and liquid lipid. The block co-polymer is selected from the group comprising polyethylene glycol-polylactide (PEG-PLA), and poloxamer, poly(lactic-co-glycolic acid) (PLGA). The solid lipid is selected from the group comprising a mixture of palmitic acid ester, glyceryl monostearate, glyceryl dibehenate, and stearic acid ester. The liquid lipid is selected from the group comprising linoleic acid, oleic acid, glyceryl caprylate, and esters of glyceryl and polyethylene glycol. The invention also relates to a method of preparing the said composition.
