Abstract: ABSTRACT CLICK CHEMISTRY-ENGINEERED GOLD QUANTUM DOT-BASED FLUORESCENT MOLECULAR PROBE FOR IMAGING AND TARGETED THERAPY The present invention provides a fluorescent molecular probe system (100) comprising gold quantum dots functionalized with amikacin via EDC/NHS coupling chemistry for theranostic applications against multidrug-resistant ESKAPE pathogens. The probe is synthesized through a click chemistry approach in aqueous medium using chloroauric acid reduction with tri-sodium citrate followed by L-cysteine surface passivation to generate fluorescent gold quantum dots exhibiting spherical morphology (6 plus or minus 3 nm diameter), negative zeta potential (minus 21.06 mV), and strong photoluminescence (excitation 420 nm, emission 497 nm). The GQD-amikacin conjugate demonstrates dramatically enhanced antimicrobial sensitivity (1300-fold and 520-fold against K. pneumoniae and A. baumannii respectively), effective biofilm inhibition at sub-MIC concentrations, capsule reduction, increased membrane permeability, and successful intracellular targeting with macrophage internalization. The dual diagnostic-therapeutic capability positions this invention as a theranostic platform for combating drug-resistant infections. [
