Abstract: ABSTRACT CLOSED-LOOP PERFUSION CONTROL SYSTEM FOR MULTI-ORGAN MICROFLUIDIC PLATFORMS A closed-loop perfusion control system for multi-organ microfluidic platforms comprises a plurality of organ modules (10) comprising interconnected patient-derived organ compartments configured to receive a flow of culture media, a micro-electrode sensor array (20) coupled to the plurality of organ modules (10) and configured to measure physiological parameters comprising pH, dissolved oxygen, and trans-endothelial electrical resistance from the organ compartments, an embedded controller (30) communicatively coupled to the micro-electrode sensor array (20), a serial bus transceiver (40) coupled to the embedded controller (30) and configured to transmit flow setpoint commands, and an electromechanical micropump (50) coupled to the serial bus transceiver (40) and configured to regulate the flow of culture media through the plurality of organ modules (10) in response to the flow setpoint commands. The embedded controller (30) is configured to normalize sensor readings to a per-organ metabolic-demand vector, compute a per-organ target flow with a shear penalty that suppresses flow as predicted wall shear stress approaches a per-tissue shear ceiling, and resolve conflicting per-organ target flows into a consensus flow bounded by a safe-flow ceiling of a most fragile organ compartment.
