Abstract: The present invention relates to the development of a CRISPR-Cas9 genome editing system to confer resistance against Cotton leaf curl Multan virus (CLCuMuV) inducing cotton leaf curl disease (CLCuD) in G. hirsutum cultivar HS6. The invention discloses the design and cassette of multiplex single guide RNA (sgRNA) cassette targeting overlapping (C1/C4) regions of CLCuMuV in sense and antisense orientations. Transgenic cotton plants, cells, or tissues expressing the CRISPR-Cas9 system are generated through Agrobacterium-mediated transformation. Upon viral inoculation, the Cas9/sgRNA complex induces site-specific double-strand breaks (DSBs) in the viral DNA, leading to disruption of viral replication. The invention provides a durable genome-editing-based strategy for the developing begomovirus-resistant cotton cultivar and establishes a broadly applicable antiviral platform for crop improvement.
