Abstract: A method for CRISPR-Cas9 gene editing comprising introducing a CRISPR-Cas9 system, comprising a Cas9 enzyme and a guide RNA (gRNA) specifically designed to target the INS gene, into pancreatic beta cells. It is delivered using either viral vectors, such as adeno-associated viruses (AAV), or non-viral vectors, such as lipid nanoparticles. The Cas9 enzyme induces a double-strand break at precise location within the INS gene, activating the cell's natural repair mechanisms, including non- homologous end joining (NHEJ) or homology-directed repair (HDR), to restore the gene's function. The method can be tailored to the patient's specific INS gene mutation or defect, and may include a gene repair or replacement template to facilitate precise genetic corrections. The CRISPR-Cas9 system can be delivered in vivo or ex vivo, with the latter involving reintroduction of the edited cells into patient. This approach is optimized for each patient's unique genetic makeup to achieve sustained endogenous insulin production
