Abstract: The present invention relates to a lyophilized liposomal Amphotericin B formulation optimized using Box–Behnken experimental design. The formulation comprises Amphotericin B, hydrogenated soy phosphatidylcholine, cholesterol, and trehalose. HSPC forms a rigid liposomal bilayer for incorporation of Amphotericin B, cholesterol stabilizes the bilayer and reduces drug leakage, and trehalose protects the vesicles during freezing, drying, storage, and reconstitution. The formulation is prepared by lipid film formation, hydration, vesicle size reduction, trehalose addition, and lyophilization. Box–Behnken experimental design is used to optimize HSPC, cholesterol, and trehalose concentrations with respect to particle size, polydispersity index, entrapment efficiency, reconstitution time, leakage, hemolytic toxicity, and antifungal performance. The optimized formulation provides high Amphotericin B entrapment efficiency, controlled particle size distribution, reduced hemolytic toxicity, improved storage stability, rapid reconstitution, and improved systemic antifungal performance for the treatment of systemic fungal infections.
