Abstract: The present invention relates to a novel method for preparing sustained-release oral pharmaceutical tablets using in situ forming polyelectrolyte complexes (PECs) composed of chitosan and natural anionic polymers such as gum ghatti or xanthan gum. The method involves preparing cooled, neutralized polymeric binder solutions which are used in a wet granulation process to formulate high-dose, water-soluble drugs like paracetamol. Upon exposure to acidic gastric conditions, these polymers interact to form a gel-like PEC barrier that retards drug release. The resulting tablets demonstrate controlled zero-order release over extended periods, improved mechanical properties, and reduced burst effect. This invention provides a simple, scalable, and biocompatible approach for sustained drug delivery without the need for synthetic crosslinkers, offering broad applicability for drugs with short halflives and high solubility.
