Abstract: Development of Rapid Disintegrating Quercetin Tablets Using Solid Dispersion and Super-Disintegrants This invention describes the development of rapid disintegrating tablets (RDTs) for Quercetin, a poorly water-soluble flavonoid with therapeutic potential. Utilizing solid dispersion techniques with Poloxamer 188 as a hydrophilic carrier and optimized super-disintegrants such as cross-povidone and croscarmellose sodium, the formulation significantly enhances solubility, dissolution, and bioavailability. The solid dispersion, prepared using the solvent evaporation method at a 1:3 drug-to-polymer ratio, ensures uniform drug distribution and improved dissolution rates. The super-disintegrants, optimized at 8% concentration, enable rapid tablet disintegration within 23 seconds upon contact with saliva. This ensures 98.15% drug release within 60 minutes, facilitating faster therapeutic action. The formulation is user-friendly, requiring no water for administration, making it ideal for populations with swallowing difficulties. Scalable for industrial production and validated for stability, this invention offers a robust solution for overcoming bioavailability challenges associated with poorly soluble drugs, ensuring enhanced patient compliance and therapeutic efficacy.
