Abstract: ABSTRACT TITLE OF THE INVENTION: Diploid yeast engineering method and strain development for co-utilization of glucose-xylose from lignocellulosic biomass A genomically modified diploid Lachancea kluyveri strain capable of co-utilization of glucose and xylose during fermentation harbored heterologous xylose metabolic pathway genes with upregulated transaldolase and transketolase. In bioreactor fermentation, the recombinant host utilized glucose and xylose, obtaining ethanol yield without xylitol accumulation. Further, fermentation using biomass hydrolysate resulted in complete consumption of xylose to produce ethanol. Higher expression of XYL2 with overexpressed TAL, TKL eliminated xylitol accumulation completely in engineered L. kluyveri. Heterologous genes XYL1, XYL2, XYL3 were expressed under the identified strongest promoter PTEF1. Chromosomal integration of genes at both the alleles of diploid L. kluyveri enabled stable expression suitable for industrial applications. Large scale bioreactor fermentation showed ethanol yield of 0.42 g/g sugar with a high total sugar consumption rate of 1.57 g/L/h by simultaneous utilization of glucose and xylose Fig. 1
