Abstract: The present invention discloses a dual reactive oxygen species and pH responsive smart hydrogel adhesive designated DTQO@TS/KM for tendon-bone interface regeneration. The hydrogel comprises tannic acid-strontium metal-phenolic nanoparticles and kartogenin-loaded mesoporous polydopamine nanocarriers within a triple- crosslinked network formed by thioether bonds, boronate ester bonds, and dynamic imine bonds via photo-crosslinking of polyethylene glycol diacrylate, dithiothreitol, boronic acid-modified quaternized chitosan, and oxidized dextran. The triple-crosslinked architecture provides dual ROS and pH responsive drug release, with cumulative release reaching approximately eighty percent under pathological conditions. The hydrogel adhesive demonstrates compressive modulus exceeding seven hundred kilopascals, ABTS radical scavenging exceeding ninety-seven percent, and promotes macrophage M2 polarization, osteogenic differentiation, and chondrogenesis. Animal studies confirm enhanced collagen remodeling, fibrocartilage regeneration, and improved biomechanical strength at the tendon-bone interface. The invention provides a cost-effective therapeutic platform utilizing abundantly available tannic acid for rotator cuff repair.
