Abstract: The present invention relates to an eco-friendly staining method for nucleic acids using walnut husk extract. The method includes preparation of a walnut peel extract solution, electrophoretic separation of nucleic acids on an agarose gel, washing of the gel, staining by immersing the gel in the extract solution for 30 to 60 minutes, and destaining using a mixture of acetic acid, methanol, and water until distinct dark brown DNA bands are visible. The extract shows affinity towards nucleic acids, allowing visualization of DNA bands with the naked eye without the use of a UV transilluminator. The staining solution (solution of walnut husk extract) can be recovered and reused. The method provides a safer and environmentally benign alternative to conventional nucleic acid stains.
