Abstract: ABSTRACT ENGINEERED MULTI-PATHWAY SORTING-MOTIF EXOSOME CASSETTES FOR CARTILAGE REGENERATION AND THICKENING The instant disclosure provides a regenerative cartilage concoction comprising thirty engineered synthetic genetic expression cassettes classified into cytoprotective, immune-modulation, partial-reprogramming, chondrogenic SOX, anabolic ligand, growth-oriented receptor, matrix-organisation, pathway-antagonist and anti-fibrotic modules, the cargo of which is loaded into or displayed upon engineered extracellular vesicles by a multi-pathway sorting-motif architecture comprising C1C2, Syntenin-1, NDFIP1, DLG4, mir-1289 or hnRNPA2B1 and motifless formats. A partial-reprogramming sub-module is delivered as Syntenin-1 luminal cargo through a prolyl-endopeptidase and thimet-oligopeptidase cleavable linker, each anabolic ligand is co-presented with its cognate receptor, and cargo is constitutively expressed on a self-inactivating lentiviral backbone bearing an inducibly-gated antibiotic-selection marker. The instant disclosure further provides a zone-resolved bio-fabricated hydrogel micro-thread scaffold article incorporating the engineered extracellular-vesicle population along a superficial-to-deep spatial gradient and a dual-wave temporal release sequence, in a first configuration for focal osteochondral defects and a second configuration for diffuse osteoarthritic thinning, and methods of manufacture thereof.
