Abstract: A bio-responsive anti-inflammatory hydrogel composition (100), configured for site-specific and inflammation-responsive therapeutic delivery. The hydrogel composition (100) comprises a polymeric scaffold (102) formed from a biocompatible hydrophilic polymer, an inflammation-sensing crosslinking polymer (104) comprising reactive oxygen species (ROS)-cleavable linkages, and a pharmaceutical payload moity (106) containing one or more anti-inflammatory, immunomodulatory, or biologically active agents. A self-regulated degradation polymer (108), structurally defined by the ROS-cleavable linkages, enables selective degradation of the hydrogel in response to elevated ROS levels characteristic of pathological inflammatory microenvironments. A localized release control moity (110) is functionally coupled to the degradation polymer to regulate therapeutic release proportionally to ROS-induced cleavage. The method of manufacture comprises forming a crosslinked hydrogel network and configuring ROS responsiveness through controlled crosslinker composition and distribution. The invention provides autonomous, localized, and controlled therapeutic delivery while maintaining stability under normal physiological conditions.
