Abstract: The present disclosure relates to microbial cells, engineered enzymes, and methods for producing mogrol or mogroside. The method comprises providing a microbial cell producing squalene and expressing a biosynthetic pathway that converts squalene to the mogrol or mogroside and culturing the microbial cell under conditions suitable for producing the mogrol or mogroside. In various embodiments, the methods disclosed herein produce squalene derivatives with less than about 40% by weight of off-pathway products cucurbitadienol, hydrolyzed 2,3;22,23-dioxidosqualene, and glycosylated 24,25-dihydroxycucurbitadienol.
