Abstract: Title: “Ethanol-Mediated One-Pot Synthesis of 3-Phenyl-3,4,5,12-tetrahydrobenzo[4,5]imidazo[2,1-b]quinazolin-1(2H)-ones as PDGFRA Inhibitors” ABSTRACT: The present invention relates to an ethanol-mediated one-pot synthetic methodology for the preparation of novel 3-phenyl-3,4,5,12-tetrahydrobenzo[4,5]imidazo[2,1- b]quinazolin-1(2H)-one derivatives. The synthesized compounds exhibit potent inhibitory activity against platelet-derived growth factor receptor alpha (PDGFRA), a member of the type III receptor tyrosine kinase family, which is implicated in the pathogenesis of various malignancies. This streamlined and environmentally benign synthetic approach enables efficient access to pharmacologically relevant heterocycles with potential application in targeted cancer therapy.
