Abstract: The present invention discloses an antidiabetic pharmaceutical composition comprising a standardized ethanolic leaf extract of Gymnema sylvestre enriched in gymnemic acids and a method for evaluating its antidiabetic activity in a streptozotocin-induced diabetic rat model. The extract, standardized to contain not less than twenty-five percent gymnemic acids, was administered orally at 200 and 400 milligrams per kilogram body weight for twenty-eight days to streptozotocin-induced diabetic Wistar rats. The composition demonstrated significant dose-dependent reduction in fasting blood glucose by fifty-five to sixty-five percent, reduction in glycosylated haemoglobin, restoration of serum insulin levels, and amelioration of diabetic dyslipidaemia. Histopathological examination confirmed pancreatic beta-cell regeneration in treated animals. The higher dose exhibited efficacy comparable to glibenclamide, the standard drug. Acute toxicity evaluation confirmed safety with LD50 exceeding 2000 milligrams per kilogram. The composition provides an effective, safe, and economically viable plant-derived alternative for management of Type 2 diabetes mellitus.
