Abstract: [049] The present invention relates to an extended-release oral matrix tablet composition of atorvastatin calcium and to a process for preparation thereof. The composition comprises atorvastatin calcium or a pharmaceutically acceptable form thereof dispersed in a polymeric release-modifying matrix containing one or more polymers selected from xanthan gum, povidone, hydroxypropyl methylcellulose, and combinations thereof, optionally together with conventional pharmaceutical excipients. The formulation is designed to provide controlled release of atorvastatin over an extended period, preferably up to about 12 hours, while maintaining acceptable pre-compression and post-compression properties, including granule flow, tablet hardness, friability, thickness, and drug content uniformity. The composition may be prepared by wet granulation followed by drying, lubrication, and compression. Compatibility and stability studies indicate that the selected excipient system is suitable for atorvastatin matrix tablet development.
