Abstract: The present invention provides a pharmaceutical composition comprising a self-assembled ferritin nanocage of approximately twelve nanometres outer diameter, within which is encapsulated a curcumin prodrug. The said curcumin prodrug comprises a curcumin moiety covalently bonded through a stimulus-cleavable linker, selected from a disulfide bond, an ester bond, or a peptidic-amide bond, to a hydrophobic auxiliary moiety. The said hydrophobic auxiliary moiety non-covalently anchors the prodrug to the inner surface of the ferritin nanocage, preventing premature payload release in systemic circulation and stabilising reassembly of the nanocage. The composition affords active tumor targeting through native binding of ferritin to transferrin receptor type-1, dual stimulus-responsive intracellular release triggered by elevated tumor-cell glutathione or esterase activity, drug loading exceeding eighty-five percent and narrow particle size distribution.
