Yadav; Dr. Parveen Kumar; Dr. Rajesh Verma; and Dr. Himmat Singh Chawra on June 13, 2026, for Formulation And Evaluation Of A Bilayer Tablet Of Ketoprofen And Omeprazole Using Box–behnken Design.

Abstract: The present disclosure provides systematically formulated and evaluated bilayer tablet of Ketoprofen and Omeprazole for combination therapy. Bilayer tablet is for immediate release of Ketoprofen for rapid analgesic and anti-inflammatory effect and delayed or controlled release of Omeprazole for gastric protection. Box – Behnken Design (BBD) was used to optimize the formulation to study the effect of critical formulation variables such as polymer concentration, excipient ratios and compression force on the performance of tablets. The bilayer tablet is manufactured using controlled blending, granulation and sequential compression processes to achieve the distinct layers with minimal intermixing. The formulated product exhibits improved mechanical strength, stability and reproducible drug release profiles. The in vitro evaluation confirms the rapid disintegration of the Ketoprofen layer and the effective protection and release of Omeprazole under intestinal conditions. The system described provides improved therapeutic efficacy, reduced gastrointestinal side effects and improved patient compliance compared to conventional single layer dosage forms.
