Abstract: The present invention relates to a cisplatin loaded gum ghatti nanoparticles prepared by nanoprecipitation method. The nanoparticulate composition, comprises cisplatin entrapped within a gum ghatti polymeric matrix optionally in presence of surfactant. The optimized formulation, comprises 100 mg cisplatin, 150 mg gum ghatti polymer, 100 mg surfactant, 20 ml aqueous solvent and 10 ml organic solvent. The process for preparation of cisplatin loaded gum ghatti nanoparticles comprising: preparing cisplatin solution in water using DMSO as co-solvent; dissolving gum ghatti in acetone to form polymeric organic phase; mixing the organic phase with cisplatin solution to form dispersion; stirring the dispersion; adding the organic phase into aqueous phase; evaporating solvent; centrifuging the formed nanoparticles; washing and drying the nanoparticles. The prepared nanoparticles exhibited average particle size of 143.4 nm, polydispersity index of 0.160 and zeta potential of +28.5 mV. The nanoparticles further exhibited encapsulation efficiency of 93.56%, drug loading of 83.55% and percentage yield of 76.54%. In vitro dissolution studies using USP Apparatus II in phosphate buffer pH 6.8 at 37±0.5°C and 50 rpm demonstrated about 98.53% drug release within 120 minutes. The formulation further demonstrated anticancer, immunomodulatory and antimicrobial activity in biological evaluations.
