Abstract: The present invention discloses topical cream formulations comprising ethanolic, aqueous, and hydroalcoholic extracts of Bidens pilosa L. (Asteraceae) incorporated at 5 percent w/w in oil-in-water cream bases, and a systematic method for evaluating their dermal safety. Physicochemical evaluation confirmed acceptable pH (6.35 to 6.58), viscosity (11,800 to 12,450 cP), spreadability, and drug content (96.5 to 98.1 percent). Acute dermal irritation studies per OECD 404 demonstrated primary irritation indices of 0.50, 0.17, and 0.83 for ethanolic, aqueous, and hydroalcoholic extract creams respectively, all classified as negligible irritants. Sub-acute 28-day repeated application per OECD 410 revealed no mortality, no systemic toxicity, normal hematological and biochemical parameters, and unremarkable skin histopathology. Skin sensitization studies per OECD 406 showed 0 percent sensitization for all formulations. The aqueous extract formulation exhibited the best dermal safety profile. The invention provides safe, naturally derived topical alternatives for dermatological applications.
