Abstract: ABSTRACT TITLE: FORMULATION OF INTRANASAL NANOEMULSIONS FOR ENHANCED BRAIN TARGETING IN ALZHEIMER’S DISEASE The present invention discloses an intranasal nanoemulsion formulation for enhanced brain targeting in Alzheimer's disease comprising a lipid-based oil phase containing an active pharmaceutical ingredient, a surfactant-co-surfactant blend, an aqueous phase, a mucoadhesive polymer coating layer comprising chitosan, and an in situ gelling module comprising a thermosensitive poloxamer. The nanoemulsion globules have a mean diameter of 20 to 200 nanometers and exhibit a positive zeta potential ensuring mucoadhesion to nasal mucosa. Upon intranasal administration the formulation undergoes sol-to-gel transition at nasal temperature forming a sustained-release depot. The active ingredient is transported to the brain via olfactory and trigeminal nerve pathways bypassing the blood-brain barrier. The formulation achieves enhanced brain bioavailability, reduced systemic exposure, and is non-toxic to nasal tissue.
