The application for the patent was published on October 30, 2026, under issue no. 44/2026.

Abstract: The present invention discloses a gastroretentive effervescent bilayer tablet for oral semaglutide delivery comprising a first effervescent floating layer containing sodium caprate as a permeation enhancer within an HPMC K4M gel-forming sustained-release matrix, with sodium bicarbonate and citric acid as gas-generating agents, and a second drug-bearing layer containing enteric-microcoated semaglutide granules coated with HPMC-AS. Upon ingestion, carbon dioxide entrapped within the gel matrix provides buoyancy within three minutes, maintaining flotation for more than four hours. Controlled sodium caprate release raises local pH above 5.5, enabling dissolution of the enteric microcoating and releasing semaglutide into the enhancer-enriched gastric microenvironment. In beagle dogs, the formulation achieves 160.6% relative bioavailability versus the commercial reference product, with coefficient of variation of 75.4% versus 139.8%. The invention resolves the prior-art conflict between enteric protection and gastric absorption by selectively microcoating peptide granules, providing enhanced oral peptide delivery and industrial applicability.
