Abstract: The present invention discloses glytrexate-loaded pH and reactive oxygen species dual-responsive polymersomes for breast and solid tumor therapy. The polymersomes are self-assembled vesicles formed from an amphiphilic methoxy poly(ethylene glycol)-thioketal-poly(beta-amino ester) (mPEG-TK-PBAE) triblock copolymer, encapsulating the hydrophilic multitargeted antifolate glytrexate within an aqueous lumen surrounded by a stimuli-cleavable polymeric bilayer. The polymersomes have a particle size of 80 to 150 nanometers, encapsulation efficiency above 65 percent, and remain stable at physiological pH 7.4 while undergoing rapid disassembly under combined tumor-microenvironment cues of mildly acidic pH and elevated hydrogen peroxide. The invention provides four-fold lower half-maximal inhibitory concentration in 4T1 breast cancer cells, prolonged plasma half-life, 76 percent tumor volume reduction in vivo, 82 percent reduction in lung metastasis, and markedly reduced systemic toxicity compared to free glytrexate and
