Abstract: [049] The present invention relates to a green synthesized metallic nanoparticle in situ gel composition comprising Lawsonia inermis leaf extract for neuroprotective delivery associated with Alzheimer’s disease. The leaf extract functions as a natural reducing, capping, stabilizing, and bioactive phytochemical source for preparing metallic nanoparticles under aqueous green synthesis conditions. The nanoparticles are incorporated into a physiologically responsive in situ gel matrix comprising gelling and mucoadhesive polymers. The composition remains flowable before administration and forms a gel under nasal physiological conditions, thereby increasing nasal residence time, reducing rapid clearance, and providing sustained release of phytochemical-capped metallic nanoparticles. The formulation supports brain-directed delivery through nasal mucosal pathways and may provide antioxidant, anti-inflammatory, and neuronal protective benefits relevant to Alzheimer’s disease-related neurodegeneration. The invention also provides a process for preparing the composition through leaf extraction, green nanoparticle synthesis, nanoparticle purification, gel base preparation, and incorporation of nanoparticles into the in situ gel matrix.
