Abstract: ABSTRACT OF INVENTION HERBAL ANTI-DIABETIC TABLET AND ITS PROCESS OF FORMULATION This invention presents an innovative herbal antidiabetic formulation using water-soluble extracts from Neolamarckia cadamba and Swertia chirayita. The formulation aims to provide an effective, cost-efficient treatment for diabetes and its complications, including nephropathy and retinopathy. The formulation is designed as a sustained-release herbal tablet, evaluated for entrapment efficiency, drug release, and hardness. In vitro release and ex vivo diffusion studies revealed that Mangiferin (from Swertia chirayita) released 79.45%, and Quercetin (from Neolamarckia cadamba) released 80.80%, showing optimal drug release kinetics. Comparisons with plain drug solutions demonstrated the advantages of the formulated dosage form. A Design of Experiments (DoE) approach was used to optimize the formulation. Thirteen batches were prepared using a Quadratic design with HPMC K4M and Ethyl cellulose concentrations as independent variables, and tablet hardness, %CDR1, and %CDR2 as responses. Based on this analysis, the final formulation demonstrated desirable characteristics. Stability studies under ICH guidelines confirmed the formulation’s stability, showing no significant changes after three months under both intermediate and accelerated conditions.Animal studies on Wistar rats evaluated the formulation’s efficacy. Rats were divided into four groups: Group 1 (Normal control), Group 2 (Disease control with STZ-induced diabetes), Group 3 (Sitagliptin treatment), and Group 4 (Formulation treatment at 260 mg/kg body weight). Over 28 days, Group 4 showed significant antidiabetic effects, improving glycemic control and insulin sensitivity. This herbal formulation offers a safer, cost-effective alternative to conventional antidiabetic treatments. It provides a holistic, single-tablet solution combining the therapeutic properties of Neolamarckia cadamba and Swertia chirayita, offering an economically viable option for managing diabetes and its complications with minimal side effects compared to synthetic drugs.
