Abstract: HPLC METHOD FOR DETECTION OF ESOMEPRAZOLE MAGNESIUM TRIHYDRATE BP AND LEVOSULPIRIDE CAPSULE DOSAGE FORM The present invention relates to a novel High-Performance Liquid Chromatography (HPLC) method for the simultaneous estimation of Esomeprazole Magnesium Trihydrate BP and Levosulpiride in capsule dosage form, addressing the need for accurate, cost-effective, and rapid routine analysis. Each capsule contains 40 mg of Esomeprazole and 75 mg of Levosulpiride, which are used in managing gastrointestinal conditions such as GERD and ulcers. The method employs a mobile phase of acetonitrile and phosphate buffer (pH 7.4) in a 35:65 ratio, a stainless steel column (25 cm x 4.6 mm) with octadecylsilane-bonded silica (5 µm), a flow rate of 1.0 ml/min, and UV detection at 290 nm. Standard and test solutions are prepared using methanol, diluted with the mobile phase, and filtered. Quantification is performed by injecting 20 µL of these solutions into the HPLC system and using specified formulas based on chromatographic responses, weight, and potency of the drugs. This method is simple, precise, reproducible, and suitable for routine quality control in pharmaceutical manufacturing.
