Abstract: The present invention discloses hyaluronic acid-coated bilosomes comprising ceramide and sodium taurocholate for buccal drug delivery. The bilosomes are prepared by thin film hydration wherein ceramide, cholesterol, and sodium taurocholate form nanosized vesicles subsequently coated with hyaluronic acid through electrostatic adsorption. The ceramide bilayer biomimetically integrates with the buccal epithelial lipid barrier to facilitate transcellular permeation. Sodium taurocholate transiently modulates desmosomal junctions for enhanced paracellular transport while exhibiting reduced cytotoxicity compared to dihydroxy bile salts. The hyaluronic acid coating imparts mucoadhesion via CD44 receptor interaction, prolonging buccal residence time. The bilosomes exhibit particle size below 200 nm, high colloidal stability across physiological pH, and maintain cell viability above 80 percent at therapeutic concentrations.
