MUMBAI, India, Nov. 2 -- Intellectual Property India has published a patent application (202641120601 A) filed by Srinivasni H; Francis Saleshier M; Dr. Jayaprakasam R; and Dr. Sriram S on October 08, 2026, for In Silico And Experimental Approach For Design, Synthesis And In Vitro Evaluation Of Isatin-Based Acetylcholinesterase Inhibitors.
Inventors include Srinivasni H; Francis Saleshier M; Dr. Jayaprakasam R; and Dr. Sriram S.
The application for the patent was published on October 30, 2026, under issue no. 44/2026.
Abstract: The present invention relates to novel isatin-based Schiff-Mannich base derivatives of formula (1), being 1-[(NR1R2)methyl]-3-(arylimino)-1,3-dihydro-2H-indo1-2- ones, wherein Ar is pyridin-4-yl, pyridin-3-y1 or 3-chloro-4-fluorophenyl and N12112.2 is piperidin- 1 -yl, pyffolidin- 1 -yl, morpholin-4-y1 or dimethylamino, as acetylcholinesterase inhibitors for Alzheimer's disease. The compounds are identified by an integrated process of network pharmacology, virtual screening, drug- likeness and ADMET profiling, molecular docking and molecular dynamics, and are prepared by condensing isatin with an aromatic amine to give a Schiff base, followed by aminomethylation with formaldehyde and a secondary amine The compounds show concentration-dependent inhibition of acetylcholinesterase in Ellman's assay; 34(3-chloro-4-fluorophenyl)imino]-1-[(dimethylamino)methyl]- 1,3-dihydro-2H-indol-2-one shows an IC50 of 56.33 ug/mL, comparable to donepezil (53.6 pig/mL).
