Abstract: The present invention discloses optimized in-vitro propagation method for enhancing Diosgenin production in Costus speciosus and a method of quantification using chromatographic techniques. In one aspect, the present invention provides an efficient protocol for in-vitro propagation and Diosgenin production from Costus speciosus. In another aspect, the present invention provides a method, the method comprising: a) explant collection and surface sterilization, b) shoot induction, c) shoot multiplication, d) Diosgenin production, and e) quantification of produced Diosgenin. In another aspect, the present invention provides a quantification method for quantifying Diosgenin obtained by in-vitro propagation, the method comprising employing TLC and HPLC analytical techniques.
