Abstract: [0080] The present invention relates to a silymarin-embedded nanostructured lipid carrier (NLC) based transdermal drug delivery system and methods of preparation and use thereof. The nanostructured lipid carrier comprises a solid lipid, a liquid lipid, a surfactant, and a phospholipid, wherein silymarin is encapsulated within the lipid matrix in an amorphous or molecularly dispersed form. The NLCs are incorporated into a polymeric transdermal patch to facilitate enhanced skin permeation, controlled drug release, and improved bioavailability. The NLC-loaded transdermal patch demonstrates sustained release of silymarin and significantly enhanced ex vivo skin permeation compared to conventional silymarin patches. The invention provides a non-invasive and effective platform for transdermal delivery of poorly water-soluble therapeutic agents.
