Abstract: ABSTRACT Intranasal Thermoresponsive Formulation Incorporating IgY Antibodies for the Prevention and Treatment of SARS-CoV-2 Infection The present invention is directed towards the preparation of a thermoresponsive intranasal pharmaceutical formulation that incorporates immunoglobulin Y (IgY) antibodies specifically generated against severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). The IgY antibodies were extracted and purified from the egg yolks of hens that had undergone hyperimmunization with the receptor-binding domain (RBD) of the spike protein associated with SARS-CoV-2. The formulation includes a thermoresponsive polymer, preferably Poloxamer 407, which undergoes a sol-to-gel (solution to gel form) transition at the physiological temperature of the nasal mucosa. This characteristic prolonged the residence of antibodies incorporated in the nasal spray on the nasal mucosa, resulting in improved bioavailability. Furthermore, the invention included processes for antibody production, formulation approaches, and therapeutic applications aimed at preventing or reducing viral load in subjects at risk of, or affected by, SARS-CoV-2 infection.
