Abstract: The present invention discloses an eco-friendly and cost-effective method for the isolation, screening, and characterization of oil-degrading bacteria from petroleum- contaminated soil collected from petrol stations and related sites. The isolated bacterial strains are enriched and screened using Mineral Salt Medium (MSM) containing petroleum hydrocarbons as the sole carbon source. These microorganisms possess the ability to utilize and degrade hydrocarbons into simpler and less toxic compounds through enzymatic biodegradation. The isolated strains are further characterized using colony morphology, Gram staining, and biochemical tests for identification. The developed method provides a sustainable biological approach for the remediation of petroleum-contaminated soil and wastewater, reducing environmental pollution and promoting ecosystem restoration. The invention offers a simple, efficient, and environmentally friendly alternative to conventional remediation techniques.
