Abstract: The present invention relates to a method for the extraction and isolation of short chain fatty acids (SCFAs) from bacterial cultures, which involves a series of steps to efficiently separate and purify SCFAs for downstream analysis and characterization. The method begins with the preparation of a bacterial culture in a broth medium, followed by the separation of bacterial cells from the broth through centrifugation to obtain a supernatant containing SCFAs. The supernatant is then acidified with 1M hydrochloric acid to adjust the pH to 2.5, protonating the SCFAs, and sodium chloride is added to the acidified supernatant. Ethyl acetate is introduced as an organic solvent, and the mixture is vortexed to partition SCFAs into the ethyl acetate layer. The organic layer containing SCFAs is collected and subjected to rotary evaporation under reduced pressure to concentrate the SCFAs.
