Abstract: The present invention relates to a method for the sustainable production and application of fungal xylanase in biomass valorization, specifically focusing on the isolation and identification of fungal strains from soil samples in Punjab, India, capable of producing high levels of xylanase. The method involves cultivating the identified fungal strain in a submerged fermentation process using a liquid medium with xylan as the carbon source, and optimizing the fermentation process by adjusting parameters such as inoculum size, pH, and aeration using the One-Factor-At-a-Time (OFAT) method. Statistical techniques are employed to optimize media components for enhanced enzyme activity and protein concentration. The crude xylanase is applied to hydrolyze xylan-rich agro-residues, with enzyme activity assessed by estimating the sugar released from the biomass. The process analyzes hydrolysis with scanning electron microscopy and FTIR to observe biomass structure changes and monitors sugar release via HPLC.
