Abstract: Method of making Mucoadhesive Buccal Patch with the formulation of Metformin Hydrochloride for effective administration This invention describes a mucoadhesive buccal patch formulation of Metformin Hydrochloride, designed to enhance bioavailability and therapeutic efficacy by bypassing hepatic first-pass metabolism. Utilizing a biocompatible polymer matrix, primarily hydroxypropyl methylcellulose (HPMC K4M), the formulation ensures prolonged adhesion to the buccal mucosa and facilitates controlled drug release. The solvent casting method is employed to create uniform, flexible patches that provide consistent and sustained delivery of Metformin Hydrochloride over extended periods. The patch enhances drug permeability, enabling direct systemic absorption while minimizing gastrointestinal side effects associated with conventional oral formulations. Comprehensive evaluations, including in vitro release studies and ex vivo permeability tests, demonstrate the system's efficacy, biocompatibility, and stability. Scalable for industrial production, this invention offers a patient-friendly, non-invasive alternative for Type 2 diabetes management, improving patient compliance and therapeutic outcomes while reducing dosing frequency and adverse effects.
