Abstract: ABSTRACT MODIFIED PVA-BASED POLYMER FOR BRAIN-TARGETED CRISPR/CAS9 DELIVERY AND METHOD OF SYNTHESIS THEREOF A modified polyvinyl alcohol-based polymer for brain-targeted CRISPR/Cas9 delivery comprises a polyvinyl alcohol backbone, a first succinic acid-derived spacer connected to the polyvinyl alcohol backbone through an ester bond, phenylalanine connected to the first succinic acid-derived spacer through an amide bond, a 1,3-diaminopropane linker connected to the phenylalanine through an amide bond, a second succinic acid-derived spacer connected to the 1,3-diaminopropane linker through an amide bond, and a polyethylene glycol chain connected to the second succinic acid-derived spacer through an ester bond. The method of synthesis comprises pre-treating and activating polyethylene glycol with succinic anhydride, activating polyvinyl alcohol with succinic anhydride, conjugating with an amino acid using coupling agents, and conjugating via an amino linker. The modified polymer is formulated as a nanoformulation with CRISPR/Cas9 gene editing components for delivery to the brain.
