Abstract: ABSTRACT Modular multiplex isothermal nucleic acid diagnostic platform with disposable cartridge, integrated heating, multi-modal detection comprises a sealed disposable microfluidic cartridge (200) and a reusable device (300). The cartridge (200) contains integrated lyophilised reagents (206, 216) for nucleic acid extraction and isothermal amplification, whereby a crude sample is tested directly without separate purification and independently of a cold chain. A distribution network (208) conveys the sample to isolated reaction chambers (212, 214) for the simultaneous amplification of two or more targets, and actuated valves (210) seal the said chambers, preventing contamination and carry over. Detection zones (400) afford multi-modal detection comprising colorimetric (402), fluorescence (404) and lateral flow (406) detection. The reusable device (300) supplies isothermal heating (304), valve actuation (308) and detection (310), and operates standalone.
