Abstract: The present invention introduces a detection system that combines Loop-Mediated Isothermal Amplification (LAMP) with a trans-cleavage CRISPR method to identify the pot2 target gene from M. oryzae. Further, the invention comprises primers, guide RNA (gRNA), Cas12a protein, and a dual-labelled probe sequence. Both the isothermal amplification and the detection of target nucleic acids occur within a single reaction mixture. The samples for the detection are taken from plant tissue, and the target nucleic acid is present in quantities of fewer than 10 copies per reaction. The invention also outlines an in vitro method for detecting pot2 transposon target DNA from M. oryzae. Additionally, it includes a lateral flow assay strip kit for rapid detection of target nucleic acids in samples, alongside the LAMP-coupled trans- cleavage CRISPR-based method for identifying these nucleic acids. It is rapid, user-friendly, portable, cost-effectiveness, and suitable for field applications, while providing enhanced specificity and sensitivity.
