Abstract: The present invention relates to a mucoadhesive chitosan-coated microemulsion composition for sustained ocular delivery of brimonidine or a pharmaceutically acceptable salt thereof, particularly for the management of glaucoma and ocular hypertension. The composition comprises a drug-loaded microemulsion formed from an oil phase, a surfactant, a co-surfactant, and an aqueous phase, wherein the microemulsion droplets are further coated with chitosan to impart positive surface charge and mucoadhesive properties. The formulation is optimized to provide suitable globule size, polydispersity index, and zeta potential for ophthalmic use. The chitosan-coated system enhances precorneal retention, transcorneal permeation, and sustained release of the drug as compared with conventional eye drops. The invention further provides a method of preparation and use of the composition for prolonged intraocular pressure reduction and improved ocular bioavailability. Accompanied Drawing [FIGS. 1-11]
