Abstract: The titled invention discloses a mucoadhesive dual-action biofilm-disrupting controlled-release gel formulation for targeted treatment of chronic tonsillitis. The formulation comprises DNase (1) configured to enzymatically degrade extracellular DNA within bacterial biofilms, N-acetylcysteine (2) configured to reduce mucus viscosity and enhance penetration into biofilm-associated structures, mucoadhesive polymers (3) configured to provide prolonged adhesion to the tonsillar mucosal surface, and a controlled-release gel matrix (4) configured to provide sustained localized delivery of the active therapeutic ingredients. The integrated formulation enables synergistic disruption of persistent bacterial biofilms, improved bacterial clearance, and reduced inflammation associated with chronic tonsillitis. The localized mucoadhesive therapeutic approach minimizes systemic exposure, reduces frequent application requirements, and provides a non-invasive alternative to conventional systemic antibiotic therapy and surgical tonsillectomy for effective chronic tonsillitis management
