Abstract: ABSTRACT MULTIPLEXED BYSTANDER GENOME EDITING PLATFORM AND PROTEASE-RESPONSIVE EXOSOMAL CARGO DELIVERY SYSTEM The instant disclosure provides a coordinated genetic engineering system. A donor bystander cell construct loads Cas9 messenger RNA into vesicles for transient transfer to a target cell construct carrying a non-homologous polycistronic transfer-RNA–guide-RNA cassette directed to a fluorescent reporter and to two protease loci, whereby loss of fluorescence reports concurrent knockout and isolates a producer cell line deficient in thimet oligopeptidase and prolyl endopeptidase. An engineered construct joins an exosome-anchoring carrier protein to a macromolecular cargo through a linker matrix of 30 to 50 residues bearing cleavage motifs for both proteases; the linker remains intact in the deficient producer line and is cleaved upon exposure to functional proteases in a recipient cell, releasing the cargo.
