Abstract: The present invention relates to a multiplexed paper-based analytical device and methods for simultaneous colorimetric detection of biomarkers. The device comprises a cellulosic substrate having a hydrophobic barrier pattern defining a sample receiving region and a plurality of radially extending hydrophilic channels terminating in corresponding test zones, a plasma separation membrane laminated to a second surface of the substrate, and a plurality of reagent compositions disposed within respective test zones, each producing a colorimetric response upon reaction with a respective biomarker. The device comprises at least five detection zones configured for simultaneous detection of bilirubin, serum protein, alkaline phosphatase (ALP), aspartate aminotransferase (AST/SGOT), and alanine aminotransferase (ALT/SGPT). Methods of fabricating and using the device are also disclosed. Figure 1 is the representative figure.
