Abstract: The present invention discloses a nanoparticle-based mRNA carrier system for intranasal delivery and a method of preparation thereof. The system comprises biocompatible nanoparticles encapsulating messenger RNA and optionally modified with PEGylation and mucoadhesive agents to enhance stability, reduce immunogenicity, and improve nasal retention. The nanoparticles protect mRNA from enzymatic degradation and enable controlled release and efficient uptake through nasal mucosa. The method includes formation of nanoparticles, encapsulation of mRNA, surface modification, and formulation into an intranasal dosage form. The invention represents a novel means for delivering mRNA for the purposes of vaccines, gene therapies, and treatment without any risk of infection, providing increased bioavailability, lowered immunity response, and higher patient compliance than previous delivery systems.
