Abstract: The present invention relates to fluorinated quinoline derivatives, particularly derivatives containing a 6-fluoro-2-(trifluoromethyl)quinolin-4-yloxy)methyl moiety linked to heterocyclic systems. The derivatives comprise three series selected from 1,2,4-triazole-5(4H)-thione derivatives 5a–5d, 1,3,4-thiadiazol-2-amine derivatives 6a–6d, and 4H-1,2,4-triazole-3,4-diamine derivatives 7a–7d. In each series, the aryl substituent is selected from phenyl, 3-methoxyphenyl, 10 4-methylphenyl and 4- chlorophenyl. The derivatives are prepared by reacting 6-fluoro-2- (trifluoromethyl)quinolin-4-ol with ethyl bromoacetate, converting the resulting ester into the corresponding acetohydrazide using hydrazine hydrate, reacting the acetohydrazide with the selected aryl isothiocyanate to obtain a hydrazinecarbothioamide intermediate, and cyclizing the intermediate to obtain the respective triazolothione, thiadiazole or triazole derivative. The 15 synthesized derivatives were evaluated for anticancer; antibacterial, antifungal and anti1inflammatory activities, and selected derivatives were evaluated by molecular docking against human interleukin-1 beta
